CpG does not induce NF-κB translocation or DNA binding. A. Cells were treated with CpG, CpC, and/or IL-1β for 1 hr. Nuclear extracts were obtained, incubated with a 32P end-labeled double-stranded NF-κB oligonucleotide probe, and resolved on a gel. Unlabed NF-kB probe (comp) or AP-1 probe (non-comp) was added at 5 times the concentration of labeled probe to show specificity. This experiment was repeated twice. B. Cells were treated with CpG and/or IL-1β for 0.5, 1, or 2 h prior to extraction and resolution on a gel. Immunoblot analysis using an antibody against IκBα is shown. This experiment was repeated four times.
